If you're using the embedded ECM, you should be able to pull in the Daeja virtual viewer to accomplish this. It has the ability to load to a specific page and annotate documents. Annotations are stored as discrete objects in the ECM repository (linked to the document object) and consist of an XML that will contain the coordinates and dimensions.
I'd have to double check if there's an ootb toolkit for the viewer these days, it's probably there in the latest content toolkit, definitely if your workflow automation is case enabled.

2. Open and view ONLY chosen page from that uploaded file (normally the FIRST or LAST page) (can be edited)

4. Save the location of the rectangle to database OR a variable for integration
For the third requirement, it can be flexible, all I need is to have a rectangle on the chosen page with dimensions can be fixed.
