You should, of course, update ALL your firmware BEFORE upgrading.

When I did this to our DNS machine I had to rewrite the DNS config for named v9 - which was a learning experience but long over due as I was running v4 on AIX 5.1.

Reading the README is always a good idea!

With AIX 5.3 JFS2 and the 64 bit kernel are regarded as the normal mode s- you may want to make use of these as you upgrade.

It is also worth getting to at least AIX 5.3 ML3 rather than an older ML.

You should check you applications are supported at AIX 5.3 and if you need to update these too.

Have you a spare machine or can hire one? If yes you can mksysb clone the current configuration and try the upgrade out first. If not backup EVERYTHING TWICE before the upgrade. It is unlikely to fail but better safe than working all night.
I hope this helps, thanks N

I can't overemphasize the need to update the microcode before upgrading to AIX 5.3. You can get the microcode (and AIX 5.3 updates) at

    http://www-912.ibm.com/eserver/support/fixes/

Depending on your uptime requirements, you might want to use "alt-disk-install". It involves minimal downtime. It requires a free disk to copy/update your current OS. You'll end up with a dual boot system one for AIX 5.1 and one for AIX 5.3 so you can back out if necessary. (this doesn't eliminate the need to have a backup!! After making the backup, I'd test it. )
