If I understand it correctly, the device must be in Device Owner (Is it possible in COSU?). In order to have a device in Device Owner, a device wipe must be made.
The customer's requirement is to add devices already in use to the MDM, have separate private and business space, and be able to determine the location of the device. Is it solvable?

Not for Android Enterprise enrolled as Profile Owner (i.e. BYOD). You might choose the Device Owner enrollment for more advanced features as the whole device is a work profile.

So for phones that are in Android Enterprise there is no way to determine their location (eg in case of loss)? Or does it have to be done differently?

Location is not available on devices enrolled in Profile Owner mode with Android Enterprise. Profile owner is for BYOD, hence locating the device is not possible

I don't have the "Locate" item in the MaaS360 portal for Android devices. Location services in Android settings are enabled, and MaaS360 is allowed to use location services. Android Enterprise is enabled for Android devices.
What I need to set up to be able to find out the location of Android device in MaaS360.
